T cells undergo epigenetic changes upon T cell activation. We used the Assay for Transposase-Accessible Chromatin sequencing (ATAC-seq) to analyze epigenetic changes in T cells that were sufficient for or lacked PSGL-1 (gene: Selplg) two hours after T cell activation. Overall design: Naive PSGL-1+/+ or PSGL-1-/- OT-I CD8+ T cells were isolated using magnetic bead separation and activated using immobilized anti-CD3 antibody for 2 hours.
