Streptomyces cinnamoneus DSM 41675 is the natural producer of bicyclomycin, a broad spectrum antibiotic active against Gram-negative bacteria, and the only natural product known to target the transcription terminator Rho. This antibiotic has promising prospects, as it shows rapid bactericidal synergy when used in combination with other bacteriostatic antibiotics, and it has been shown that its potency can be increased through modification of its lateral chain. Identification of the biosynthetic gene cluster for this antibiotic will enable the engineered production of new bicyclomycin derivatives with potential clinical applications. Additionally, as a result of this study it has been found that the bicyclomycin gene cluster is unusually widespread, offering insights into the evolution and dissemination of antibiotic gene clusters and antibiotic resistance among bacteria.
