Objective To prepare monoclonal antibody to bacterial endotoxin and establish a chemiluminescence method for the quantitative detection of endotoxin.Methods BALB/c mice were immunized with bacterial endotoxin as immunogen to prepare the required monoclonal antibody. The best antibody pairs were screened, biotin and acridinesulfonamide were labeled, and the samples were quantitatively detected by automatic chemiluminescence detector. The blank limit LoB, detection limit LoD and quantification limit LoQ were evaluated and verified, and the influence of fungal (1,3) -β-D glucan on endotoxin detection was evaluated.Results The blank limit LoB was set at 0.005EU/mL, the detection limit LoD was set at 0.010EU/mL, and the limit of quantification LoQ was set at 0.020 EU/mL. Fungal (1,3) -β-D glucan had no effect on endotoxin detection.Conclusion Chemiluminescence method can be used for the quantitative detection of bacterial endotoxin.
