Gene expression profile at single cell level of CD45+ cells from the mouse melanoma after treatment with M1 or M1-mutIL-18

CD45+ immune cells play an important role in immunotherapy.To investigate the effect of CD45+ immune cells on the efficacy of oncolytic viruses M1 and M1-mutIL-18, We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of CD45+ immune cells in B16F10 tumors of mice after M1 and M1-mutIL-18 treatment. Overall design: B16F10 cells were injected subcutaneously into the right abdomen of C57BL/6J mice and treated with M1 (1.5 Ã 107 PFU) or M1-mutIL-18 (1.5 Ã 107 PFU) when the tumour volume was approximately 200 mm3 . M1 or M1-mutIL-18 was injected through the tail vein for 5 consecutive days. Mice were euthanised on the third day after the last dose and CD45+ immune cells in tumors were isolated by immunomagnetic beads (130-110-618, Miltenyi Biotec). Biological replicates from n = 3 pooled mice were processed for each experimental condition and analyzed by scRNAseq and scTCR-seq.
