Preparation of filamentous cyanobacteria for use with a droplet-based microfluidic device

This data set includes three videos of Oscillatoria acuminata collected from Port Said, Egypt. Cultures were prepared for microfluidic applications to observe its growth over time. Various filament lengths were generated (40 μm, 70 μm, and 100 μm) by sonication and captured inside 290 μm-droplets. Growth of filaments was observed for three days under different temperatures (20°C, 27°C, and 37 °C). During the observation period, the filaments were actively growing and entangled inside the droplets. The surface tension was high and contained the filaments inside the droplets without rupturing. In some cases, filaments fragmented inside the droplets to form hormogonia for continued growth.
本数据集包含三段取自埃及塞得港的尖头颤藻(Oscillatoria acuminata)观测视频。为开展微流体应用相关研究,我们制备了该藻的培养物以实时追踪其生长过程:通过超声处理制备了长度分别为40μm、70μm及100μm的藻丝,并将其封装于290μm的微液滴内。随后在20℃、27℃与37℃三种不同温度条件下,对液滴内藻丝的生长情况开展了为期三天的观测。观测期间,藻丝持续活跃生长并在液滴内部相互缠绕;液滴具备较高的表面张力,可将藻丝束缚于液滴内部而不发生破裂。部分场景下,藻丝会在液滴内部发生断裂,形成藻殖段以实现后续生长。
