Porites divaricata 2bRAD sequencing

To determine how a population of thin finger coral (Porites divaricata) living on mangrove roots at Calabash Caye, Belize reproduces and undergoes larval dispersal, we used a population genomics approach. We sequenced a small portion of the genome from 140 individual coral colonies using the 2bRAD approach. The DNA was first digested using a restriction enzyme (Bcg1), and then the restriction fragments were sequenced so that we could identify genetic variation between individuals (single nucleotide polymorphisms).
