Raw Flow cytometry dataset: Optimising Recovery of Hepatic Regulatory T Cells: A Practical Guide Using ARTC2 Blockade

Raw flow cytometry files from homeostasis and inflammation experiments +/- ARTC2 reagent fomat: raw .fcs files Acquired using a 5L Cytek Aurora Files are raw intrahepatic lymphocyte files for analysis of Treg populations in response to treatment with ARTC2 in homeostasis and inflammation. Multiple autofluorescence signatures were removed using Spectroflo software and analysis was performed in Flowjo V10.10 Aim: To identify if there are differences in the recovery of activated effector like Treg compared to less activated Treg cells from the liver in homeostasis and inflammation. Exp 1 Groups: Mouse ID numbers: 35, 38, 39 = homeostasis + no treatment Mouse ID numbers: 32, 33, 34 = homeostasis + ARTC2 reagent (50ug/mouse) i.v. 20 min prior to harvest Mouse ID numbers: 51, 52, 62, 63 = inflammation (LPS treatment 1ug/g of mouse) + no further treatment Mouse ID numbers: 48, 49, 50, 60, 61 = inflammation (LPS treatment 1ug/g of mouse) + ARTC2 reagent (50ug/mouse) i.v. 20 min prior to harvest Exp 2 Groups: Mouse ID numbers: 9, 10, 11, 12 = homeostasis + no treatment Mouse ID numbers: 13, 14, 15, 16 = homeostasis + ARTC2 reagent (50ug/mouse) i.v. 20 min prior to harvest Mouse ID numbers: 1, 3, 4 = inflammation (LPS treatment 1ug/g of mouse) + no further treatment Mouse ID numbers: 5, 6, 7, 8 = inflammation (LPS treatment 1ug/g of mouse) + ARTC2 reagent (50ug/mouse) i.v. 20 min prior to harvest
