To further explore potential molecular mechanisms and pathways by which the presence or absence of the pGKT2 plasmid may be affecting the overall fitness cost in a transconjugant Rhodococcus jostii RHA1 strain, transcriptome studies were performed. Transcriptome experiments comparing RHA1 wild-type and RHA1 transconjugant strains grown in rich media confirmed the presence of the pGKT2 plasmid. Overall design: An 8 x 15K array study using total RNA recovered from triplicate cultures of Rhodococcus jostii RHA1 wild-type strain and a RHA1 transconjugant strain that took up the pGKT2 plasmid were performed.
