SARS-CoV-2 RBD Escape Velocity Outpaces mAb Development for 2025 Variants Already Encoded in Global Diversity

Structural surveillance of 5M+ genomes and filtered 133,893 SARS-CoV-2 genomes (≤2023) reveals pervasive escape at therapeutic mAb interfaces, with 100% of sequences that have been extracted from larger dataset harboring non-reference states at ≥1 of positions 417, 484, 486, 493, 498, 501. Dominant patterns including S417N/S484A/S501H (BA.1/BA.2; 67.9K seqs), S484E/S501Y (Gamma-like; 12.3K), and S417T (Beta/Omicron convergence; 4.8K) map directly to residues 498, 505, the structurally constrained overlap zone between hACE2 and next-gen mAbs like VYD2311. Mutational entropy and lineage turnover at these sites are not stochastic, they reflect targeted selection against antibody binding, already encoded in global pre-2024 diversity. Full analysis of 10M+ genomes (2019 - August 2025) is underway, incorporating FLiRT variants (KP.3.1.1, LP.8.1, XEC) and quantifying silent pressure at 455, 456, 475, 477, 487, 489, 494 - residues poised to erode IC50 stability (0.0017 - 0.0420 μg/mL) within 12 months of market entry. Lineage extinction kinetics already outpace clinical development cycles; declining global sequencing coverage (driven by H5N1 redirection) will further probably reduce therapeutic durability assessments.
