Cells were grown to 80-90% confluency before RNA extraction. Microarray-measured basal gene expression was used to differentiate the various cell types, e.g., CAFs vs NAFs. NSCLC cells lines (H3122, PC9 and gefitinib-resistant PC9_GR), CAFs (CAF7 and CAF12), and NAFs (normal lung cancer fibroblasts, WI38, IMR90 and MRC5; normal bone marrow fibroblast, HS5) were used in the study. Secretomics, transcriptomics, and tyrosine phosphoproteomics were used to identify signaling components differentially expressed in CAFs vs NAFs and their corresponding downstream signaling effects on cancer cells.
