Long noncoding RNA NKX2-1-AS1 inhibits cell migration and regulates cell-adhesion independent of NKX2-1 in human lung carcinoma cells

The long noncoding RNA NKX2-1-AS1 is highly expressed in primary lung adenocarcinomas compared to squamous carcinomas, similar to its adjacent protein-coding gene NKX2-1, but its contribution to lung tumorigenesis is not well understood. In this study we knockdown NKX2-1-AS1 by siRNA transfection and analyze the effect on gene expression in the lung carcinoma H441 cell line. This study is comprised of two independent knock-down experiments, each containing 6 Human Gene 2.0 ST arrays profiling H441 lung carcinoma cells treated with control siRNA or a pool of three siRNAs (A, B and D) targeting the lncRNA NKX2-1-AS1 (n=3 per group in each experiment). TRIzol RNA extraction was used to prepare samples for the first experiment (Experiment 1), while spin-column RNA extraction was used to prepare samples for second experiment (Experiment 2).
