Comparision of primay murine airway basal cells and ESC-derived basal cells

The goal of this experiment is to study the transcriptomic similarity and differences between ESC-derived cultured basal cells and their primary counterparts in the same culture condition. We analyzed mouse mouse iBC culture from the directed differentiation protocol outlined in Ma et al., 2023. iBC cultures are previously sorted on Nkx2-1mCherry+/NGFR+ gate and cultured for 9 days in Basal Cell Medium. Meanwhile, we harvested airway basal cells from syngeneic adult mouse trachea and cultured them in the same BCM media. Both samples are outgrowth from NGFR+ FACS purified samples.
