Macrophages adopt mural cell function and marker expression for vascular support following ischemia

We used scRNAseq to study if perivascular macrophages adopt a mural cell identity during tissue restoration in mouse models of ischemia Overall design: A total of 755 tdTomato+PDGFRß+/- cells and 372 tdTomato-PDGFRß+ cells from ischemic muscles at day 21 post-ischemia onset, and a total 190 tdTomato+PDGFRß+/- cells and 192 tdTomato-PDGFRß+ cells from healthy muscle were FACS sorted into Smart-seq2 plates in two batches, and processed using the Smart-seq2 protocol (Picelli et al., 2013). The single-cell transcriptomic sequencing was performed at Eukaryotic Single Cell Genomics Facility at SciLifeLab, Stockholm.
