Here we investigated the transcriptomic changes induced by oleic acid treatment on human Primary Renal Proximal Tubule Epithelial Cells (RPTECs) with RNA-seq. We also profiled cells with oleic acid treatment after PLIN2 gene knockdown with siRNA treatment. Overall design: There are a total of 15 samples derived from 5 groups with 3 replicates per group. Sample 1-3: RPTECs treated with non-targeting control siRNA (NTsiRNA). Sample 4-6: RPTECs treated with NTsiRNA and processed for 6-hour oleate-BSA stimulus. Sample 7-9: RPTECs treated with PLIN2 siRNA and processed for 6-hour oleate-BSA stimulus. Sample 10-12: RPTECs treated with NTsiRNA and harvested at 2 days after 6-hour oleate-BSA stimulus. Sample 13-15: RPTECs treated with PLIN2 siRNA and harvested at 2 days after 6-hour oleate-BSA stimulus. Libraries were generated with the poly-A selection method and sequenced with the NovaSeq 6000 S4 platform (2Ã150 bp) at a target of 30 million reads per library.
