Nguni Sheep HiFi Data

The genome was sequenced from blood sample collected from Mara research station located in Limpopo province of South Africa. The research station stud herd is well connected to the national herd and registered with INTERGIS. The blood sample was collected by a trained veterinarian from a jugular vein using a needle and stored in EDTA tubes and transported on a dry ice to the lab for storage at -80. For PacBio HiFi sequencing, genomic DNA was extracted from a blood sample using Nanobind protocol for whole blood high molecular weight (HMW) DNA extraction to construct a sequencing library. The protocol was optimised for extraction from 200 μL of whole blood. Library sequencing on PacBio Sequel IIe platform was done using SMRTbell® prep kit 3.0 29 following the manufacturer’s instructions. Dovetail Omni-C library prep was performed from the same sample used for HiFi sequencing following manufacturer’s instructions.
