We enriched endothelial cells and other bone marrow cells in both fetal and adult stage to investigate Wnt signaling interaction using targeted scRNA-seq analysis. This analysis facilitate identification of sources of Wnt ligands and detection of Wnt receptor expression in bone marrow. The comparison of fetal and adult stage reveals differences of Wnt signaling in fetal and adult BM. Samples were obtained from C57/B6 wildtype mice. Endothelial cells were enriched by FACS sorting based on expression of marker CD31 and endomucin. Mature hematopoietic cells were depleted from fetal and adult bone marrow cells by lineage antibody conjugated magnetic beads. The obtained cells were directly used for targetted scRNA-seq library preparation.
