To uncover the cellular identifies of the mouse hypothalamic Lepr cells, we used single nucleus RNA-seq (snRNA-seq) from enriched hypothalamic Lepr cells. Overall design: Lepr-Cre;R26-LSL-Sun1-sfGFP mice were maintained on ad libitum chow prior to microdissection of the hypothalamus. We then isolated nuclei, FACS sorted for GFP-positive nuclei, and subjected these nuclei to snRNA-seq using the 10x Genomics 3' gene expression platform.
