We show that both parenteral recombinant FGF1 and postprandially-induced endogenous FGF1 suppress lipolytic gene expression and lipolysis in adipose tissue. During nutrient stress, such as high fat diet, this FGF1-FGFR1 circuit works both with and independently of insulin, to suppress adipose lipolysis. A single rFGF1 injection is sufficient to reduce constituitive adipose lipolysis in obese mouse models, rapidly reversing hepatic glucose production and normalizing blood glucose levels. Sustained suppression of adipose tissue lipolysis achieved with chronic rFGF1 administration leads to sustained glucose control. The identification of an adipose resident FGF1-FGFR1 anti-lipolytic cascade provides insight into both the mechanism of FGF1 control of insulin sensitivity as well the therapeutic value of rFGF1 administration in the treatment of diabetes.
