Our team has developed the Methylation-Induced Chromatin Capture (MICC)-seq technique, which takes advantage of EcoGII to label open chromatin with m6A. We use the m6A-specific digestion enzyme DpnI to
selectively recognize and digest the m6A(GATC) sites, resulting in free ends for in situ ligation downstream. By using DpnII, which selectively digests the unmethylated closed chromatin, we can bserve specific interactions within closed chromatin. This versatile method is applicable for studying various types of chromatin.
