This study investigated the immunological function of PCV2 ORF5 by ectopic expression of PCV2 ORF5 in PK15 cell line. Identifying the functional role of each PCV2 ORF associated with host cell modulation may provide better knowledge about the pathogenesis of postweaning multisystemic wasting syndrome (PMWS). PCV2 ORF5 is recently identified and the functional role of ORF5 during the pathogenesis after PCV2 infection is largely unknown. A Pig kidney epithelial cell line, PK15, was transfected with pLNCX2 (Mock-PK15) or PCV2ORF5/pLNCX2 (ORF5-PK15). Cloned PCV2 ORF5 cDNA sequence (GenBank accession FR823451.1) is derived from PCV2a type contained 180 bp encoding 59 amino acids. To identify differentially expressed mRNAs in the PCV2 ORF5 transfectant and mock, three cDNA libraries from each transfectant group (PCV2 ORF5 transfectant and mock) were generated and sequenced using the Illumina HiSeq4000 sequencer.
