The main objective of the project was to standardize the generation of primary three-dimensional (3D) cultures from spontaneous canine tumours and their use as tissue-like samples for histological, biochemical, and genomic analyses. Four tumour types were included: hemangiosarcoma, osteosarcoma, urothelial carcinoma, and oral melanoma. Growth conditions were optimized by comparing a bioreactor-based system (VITVO®, Rigenerand) with a plate-based culture system using the same scaffold employed in the VITVO® device.
Additional objectives included the assessment of tumour cell growth by comparing primary tumours and the corresponding 3D cultures in terms of immunohistochemical marker expression, mitochondrial oxidative metabolism, and next-generation sequencing (NGS) profiles.
