BACE1 role in reactive astrocytes are unknown. We used single cell RNA sequencing (scRNA-seq) to analyze reactive astrocytes in mice with and without germline Bace1. We also examine reactive astrocytes in the case of adult Bace1 conditional knockout on a 5xFAD Alzheimer's disease mouse model. Astrocytes of the Bace1-/-, Bace1+/+, 5xFAD; Bace1fl/fl, and 5xFAD; Bace1fl/fl; UBC-creER mice were isolated from brains by magnetically-activated cell sorting (MACS) according to the presence or absence of ACSA2+ and resulting cells were analyzed using scRNAseq. 3 samples of each Bace1-/- and Bace1+/+ were submitted, while 1 sample containing 3 pooled brains each from 5xFAD; Bace1fl/fl and 5xFAD; Bace1fl/fl; UBC-creER were analyzed.
