Vertebrate nervous system are composed of large number of distinct neuronal cell types. As gene expression program are likely underlying this diversity, delineating the transcriptions of individual neuronal types as critical for understanding the development and function of the nervous system. Goal: This project aims to produce a high resolution map of the vertebrate photoreceptor transcriptome. Such map would provide insights into the mechanism controlling eye development, light sensing and vision, and potentially identify therapeutic approaches to treat blinding disease. Study design: RNA-Seq is used to determine the transcriptomes of retina of wild type mice and mice that lack photoreceptor cells (Aipl1 knockout). The photoreceptor transcriptome is then identified by comparing the wild type and Aipl1 knockout transcriptomes. The RNA-Seq is carried out at depth (40 to 50 million reads per replicate in four replicates) that is sufficient to quantify the expression of transcript isoforms produced by alternative promoter use, splicing and polyadenylation.
