The endoplasmic reticulum (ER)-related bio-macromolecules play important roles in the maintenance of protein homeostasis, lipid synthesis and secretion, and trans-membrane transport of biomolecules. About one third of proteins are folded in and secreted through the ER. Many conditions, including gene mutation, aging, pathogen infection, and metabolism disorder, can lead to the accumulation of misfolded or unfolded proteins in the ER lumen, causing ER stress and activating the unfolded protein response (UPR). This study aims to elucidate the mechanism and physiological function of the protein and lipid transmembrane transport in the context of ER stress from different angles and across various scales ranging from molecules to multi organelles, cells, and tissues.
