The dataset provides research data on the effects of long-term exposure to xenobiotic cadmium and biogenic copper, administered through contaminated prey, on the protein profile of the hemolymph in Steatoda grossa (Araneae: Theridiidae) spiders. The study focuses on both quantitative and qualitative changes in the hemolymph proteome in response to artificial and natural immunostimulants: phorbol 12-myristate 13-acetate (PMA) and bacterial suspensions (Staphylococcus aureus [G+], Pseudomonas fluorescens [G-]).The collected data include:• Analysis of the protein profile in female Steatoda grossa spiders that were chronically exposed to cadmium and copper through their food and were additionally immunostimulated.• Results of heavy metal concentrations in the bodies of spiders.The protein analysis was performed using 2D DIGE. Proteins were quantified with the 2D Quant Kit and cleaned with the 2D Clean-Up Kit. They were labeled with fluorescence dyes, followed by rehydration on an IPG strip and isoelectric focusing (IEF). After focusing, proteins were reduced and alkylated. The second dimension was run using polyacrylamide gels. Protein maps were visualized with a Typhoon FLA 9500 scanner and analyzed using Image Master™ 2D Platinum 7.0 software. Spots of interest were excised after silver nitrate staining for further analysis.Peptide separation was performed using a Dionex UltiMate 3000 RSLC nano system and Bruker AmaZon Speed ETD mass spectrometer. Peptides were loaded onto a C18 precolumn, separated on a C18 analytical column, and analyzed by MS and MS/MS. Data were processed using Compass Data Analysis and ProteinScape 3. Protein identification was done via the MASCOT Server with databases restricted to Metazoa or Araneae. Proteins with a score >37 were considered identified.
