This dataset contains reference-based bulk RNA-seq (mRNA) data generated from uterine wall tissue of Holstein dairy cows in a field study of naturally occurring endometritis. The aim was to compare host transcriptional states across health, disease, and treatment conditions, and to evaluate the effect of A5B5 on inflammation-associated gene expression. Nine biological samples were included in total, with three samples per group: the A5B5-treated group (A5B5-1, A5B5-2, A5B5-3), the PBS-treated endometritis control group (unhealthy-1, unhealthy-2, unhealthy-3), and the healthy control group (healthy-1, healthy-2, healthy-3). Total RNA was extracted using TRIzol according to the manufacturer’s protocol; RNA quantity and purity were assessed using NanoDrop 2000, and RNA integrity was assessed using the Agilent 2100 Bioanalyzer. Libraries were prepared with the VAHTS Universal V10 RNA-seq Library Prep Kit (Premixed Version) and sequenced on an Illumina NovaSeq 6000 platform to generate 150 bp paired-end reads. Raw FASTQ data were processed with fastp for quality control and adapter/low-quality read removal, and clean reads were aligned to the reference genome using HISAT2. Gene-level read counts were obtained with HTSeq-count, and expression abundance was calculated (FPKM).
