Bacterial 16S V4V5 data on 4 permafrost soils inoculated with active layer soils in an aerobic incubation experiment

The study explores the role of inoculating permafrost soils with active layer microbial communities across four different permafrost types (palsa peat, sedge peat, cryoturbated soil, yedoma). Permafrost soils were either inoculated with active layer soil, a negative control (sterile H2O), or a positive control (microbial community from a temperate grassland soil). DNA was extracted with QIAGEN DNEasy PowerSoil Pro kit before incubation, one day after inoculation and after 389 days of incubation under aerobic settings at 10 degrees C. V4V5 amplicons were generated with 515f-926r primers and sequenced on MiSeq v3 2x300bp with 15% PhiX spike. Deposited amplicons were demultiplexed with Illumina BaseSpace and no other processing was carried out: sequencing adapters and nextera indices are removed, but not the primers.
