The aim of the study is to detail an approach that promotes expeditious formation of yeast artificial chromosome (YAC) candidates acting as human artificial chromosome (HAC) precursors. Rapid assembly of these precursors, coupled with efficient HAC formation driven by the spheroplast fusion and centromere seeding approach, will streamline utilization of HACs in fundamental chromosome research contexts. The sequencing data deposited here was used as part of quality control to ensure that the salient yeast strain described was/is as expected.
