In order to understand the biological role of Drosophila IRBIT, we generated null mutant fruitflies. We performed Illumina platform-based RNA-Seq analysis to investigate transcriptional consequences of the mutation using fly midguts as well as whole animals. Overall design: We performed gene expression profiling on wildtype and IRBIT mutant flies. We prepared RNA-Seq libraries from midguts and whole flies. Flies were reared to be 1 day, 7 days, 40 days, or 50 days old. Males and females separately prepared under different treatments: normal, deoxyadenosine, deoxyguanosine, hydroxyurea. We also culture flies in two different conditions of normal versus axenic conditions. Experiments were triplicated for RNA-Seq except for 50 day-old samples. We generated, stranded, single-end RNA-Seq libraries to have 50 or 76 read-lengths.
