Clostridioides difficile (C. difficile) is a Gram-positive anaerobic spore-forming bacillus widely distributed in the intestinal tracts of humans, animals and the environment. C. difficile infection is one of the main pathogens causing nosocomial infectious diarrhea. The clinical manifestations of C. difficile infection in pets are diverse, ranging from asymptomatic carriage to varying degrees of diarrhea, and it can spread widely between humans and pets, threatening human health. Understanding the carriage rate of C. difficile in canine feces is of great significance for assessing public health risks, providing scientific evidence for disease prevention and control, and safeguarding the health of both pets and humans. Such studies can also provide data support for epidemiological research, help determine the baseline prevalence of C. difficile in local pet populations in China, and offer a baseline reference for identifying potential abnormal outbreaks in the future. 1. Data Collection: A total of 500 canine fecal samples were collected from veterinary hospitals, numbered sequentially from 1 to 500, with basic information and clinical data collected and analyzed. 2. Data Detection: 16S rRNA microbial sequencing was performed on each of the 500 fecal samples, and data analysis was conducted following the standard 16S sequencing data analysis pipeline. The percentage of C. difficile detection was counted at the genus level. Samples with a detection percentage greater than 0 were marked as positive, while those with a detection percentage equal to 0 were marked as negative. 3. Data Processing: ① Count the total number of positive samples, and the average carriage rate = total positive samples / total number of samples; ② The average carrying abundance (%) = the average value of all C. difficile detection percentages; ③ Sort all C. difficile detection percentages in ascending order. Q97.5 refers to the detection value at the position of 0.975*(n+1) (rounded to the nearest integer), where n is the total number of samples. 4. Data Application: Collect the C. difficile detection percentages of the cohort samples using Python, and generate a fitted plot with Matplotlib to facilitate observation and analysis by researchers. 5. Data Classification and Grading: Evaluate the calculated C. difficile detection percentages and classify them into three categories: "high", "medium" and "low". Specifically, values greater than Q97.5 are classified as "high", values greater than 0 and less than or equal to Q97.5 are classified as "medium", and values equal to 0 are classified as "low".
