Contribution of PDGFRa Lineage cells in adult mouse hematopoiesis

Hematopoietic system is composed of various cells with a wide range of functions. Hematopoietic stem cells generate these descendant cells. Recently, it has become clear that mesodermal tissues give rise to HSCs during early development. Here, we examined the properties and functions of mesoderm-derived HSCs in the adult hematopoietic system. Overall design: We isolated mesoderm-derived HSCs using PDGFRa as a mesodermal marker. Briefly, PDGFRa lineage HPCs were isolated using PDGFRa-Cre;ROSAtdTomato mice. HSCs were isolated from bone marrow and peripheral blood. We performed single cell RNA-seq analysis on the isolated PDGFRa lineage HPCs to reveal their properties and functions.
