The expression profiles of lncRNA before and after the initiation of primordial follicle

Primordial follicle accounts for the majority of the ovarian reserve, and recent interest has focused on the mechanisms underlying the follicle initiation. LncRNA is non-coding RNA that has a pivotal role in modulating various bodily activities. Recent studies suggest that lncRNA is able to regulate gene expression at different levels and participate in myriad physiological and pathological processes. For example, lncRNA regulates differentiation of germ cell. However, the regulative effects of lncRNA on follicle initiation have not yet been systemically identified. We compared the expression profile of control group (where ovaries were obtained from three-day-yaer old mice) and experimental group (where samples were cultured in vitro for eight days) by high-throughput sequencing. Our results revealed that 3397 lncRNAs and 1990 mRNAs which showed significantly differential expression (³2.0 fold-change, P < 0.05), and these transcripts are expected to be associated with follicle initiation. Further researches are needed to uncover the mechanisms of follicle initiation based on present study. Overall design: employing high-throughput sequencing technology to describe lncRNA profiles of ovarian follicle before the initiation (3d) and the follicle after the initiation (3d ovary were cultured in vitro for 8 days)
