The aim of the experiment was to study the transcriptome changes occuring after overexpression of Erg, Fli1, Tal1, Lyl1, Lmo2, Runx1, Cbfb and Gata2 in endothelial (Endo) and enriched vascular smooth muscles (eVSM) populations. A total of 18 samples were analyzed. A2lox.i8TFs ESCs were differentiated for 3 days. Flk1+ cells were isolated and were cultured in BL-CFC culture for 1 day. Endo (CD41-/VE-cad+) and eVSM (CD41-/VE-cad-) cell populations were FACS-sorted. We collected total RNA from eVSM before hemogenic endothelium (HE) culture (t0), eVSM–dox (24h post-HE culture), eVSM+dox (24h post-HE culture), Endo before HE culture (t0), Endo–dox (24h post-HE culture) and Endo+dox (24h post-HE culture). Each sample group was analyzed by microarray using 3 biological replicates.
