Primary fixation included: 2.5 % glutaraldehyde, 2% formaldehyde in 0.1 M Na-phosphate buffer, pH 7.4. Post-fixed in 2% OSO4 in 0.1 M Na-phosphate buffer, pH 7.4. Stained en bloc in 1% uranyl acetate. The tissue was then dehydrated in a graded series of ethanol and infiltrated with Spurr's resin. Thin sections of 70 nm were trimmed using a diamond knife and post-stained in uranyl acetate and lead citrate. This micrograph image was taken using a Phillips CM 100 transmission electron microscope at an accelerating voltage of 80kV.前固定液配方为：溶于0.1M磷酸钠缓冲液（pH 7.4）中的2.5%戊二醛（glutaraldehyde）与2%甲醛（formaldehyde）。随后使用溶于0.1M磷酸钠缓冲液（pH 7.4）的2%四氧化锇（OSO4）进行后固定。采用1%醋酸铀酰（uranyl acetate）进行块染。将组织经梯度乙醇系列脱水后，以Spurr树脂（Spurr's resin）进行渗透浸润。使用金刚石刀（diamond knife）修块，切取70nm超薄切片，再以醋酸铀酰与柠檬酸铅（lead citrate）进行复染。本显微图像采用飞利浦CM 100透射电子显微镜（transmission electron microscope）拍摄，加速电压为80kV。
