The Python script used to plot Laurdan-GP values from raw fluorescent images.

The script generated for supplementary Figure 10 in publication "Insights into the regulation of VPS13 family bridge-like lipid transfer proteins from the structure of VPS13C." is used to plot Laurdan-GP values from the image dataset. The image dataset contains the two sets of raw micrographs. The experiment serves as a negative control to show that DOG lipids robustly induce packing defects on GUVs. For both POPC-GUV and POPC+DOC-GUV were treated with C-Laurdan and measured for its emission spectrum shift from I440 to I490. The shift, termed as generalized polarization, measures the local membrane packing. 20mM Claurdan_DMSO_control.czi file is the calibration file to normalize the two channels of the instrument using the GPexp=0.207.
