We employed bulk RNA-seq to find protoplasting-induced genes in the wild model grass Brachypodium distachyon. Overall design: Samples were collected from leaf developmental zones of 3 weeks old Brachypodium distachyon Bd21-3. 10 leaf developmental zones were digested to release single cells (protoplasted) and 5 leaf developmental zones were used for the non-protoplasted samples. Three replicates were produced per treatment. Total RNA extraction, poly-A-based library preparation and bulk-RNA sequencing of these samples allowed to check for genes that are affected by protoplasting.
