We used PURE-seq with single-cell RNA-sequencing (scRNA-seq) to study murine long-term hematopoietic stem cells (LT-HSCs) and their transcriptomes in the context of hematopoietic aging. Overall design: First, we used human and mouse cell mixtures to demonstrate the sensitivity and specificity of PURE-seq. We used mixed population of HEK/3T3 or 3T3/PBMC cells with target cell fractions of 10^-3, 10^-4, 10^-5, and 10^-6. Then, we used PURE-seq workflow to study murine LT-HSCs from young, middle-aged, and old mice. We also evaluated murine hematopoietic stem cells (HSCs) and the percentage of long-term HSCs (LT-HSCs) before enrichment, as a comparison to enriched LT-HSCs throuhg PURE-seq.
