Polarisation camera movie of single SYTOX Orange molecules on a cover glass

This image dataset is a movie of the fluorescence of single SYTOX Orange molecules (S34861, Invitrogen) dispersed on a cover glass. The data was collected on a fluorescence microscope (Ti-U, Nikon) with a polarisation camera (CS505MUP, Thorlabs). The molecules were excited with a 532 nm diode laser with a measured power density at the sample plane of 0.36 kW/cm^2. An exposure time of 100 ms was used. The following filters were used: dichroic (Di03-R532-t1, Semrock) and emission filter (FF01-582/64, Semrock).

本图像数据集为分散于盖玻片表面的单个SYTOX橙（SYTOX Orange，货号S34861，Invitrogen）分子的荧光时序成像序列。数据采集使用尼康（Nikon）Ti-U型荧光显微镜，搭配索雷博（Thorlabs）CS505MUP型偏振相机。实验采用532 nm二极管激光器激发样品，经实测样品平面处的激光功率密度为0.36 kW/cm²，成像曝光时长设置为100 ms。本次实验所用光学滤光元件包括：Semrock品牌的Di03-R532-t1型二向色镜，以及FF01-582/64型发射滤光片。
