Adult stem cells persist in mammalian tissues by entering a state of reversible arrest or quiescence. Here we report that promoter-proximal pausing of RNA pol II is involved in the maintenance and exit from the quiescent self-renewing state, using genome-wide ChIP-seq and RNA-seq analysis of quiescence in cultured myoblasts (C2C12 cell line). We conclude that RNA Pol II stalling mediates transcriptional repression that controls quiescence, and that G0-stalled genes contribute to self-renewal.
