Pooled Tn-Bar-Seq of Parabacteroides merdae mutant library grown on industrial and agricultural chemicals

Pooled chemical genetic screen of a barcoded transposon library from the gut bacteria Parabacteroides merdae (Tn-Bar-Seq). We used this Tn library for a fitness screen with a panel of 15 chemicals including industrial and agricultural contaminants. The chemicals TBBPA, BPAF, imazalil sulphate, fluazinam, closantel and emamectin benzoate were tested at a concentration of 20 µM or less. Glyphosate, propiconazole, PFOA and PFNA were tested at larger concentrations.
