The activity of enhancers with dynamic P300 binding or mutagenized nuclear receptor motifs was assessed by massively parallel reporter assay during CM maturation. Overall design: AAV9 reporter pools were injected at P0 or P7 and tissue was harvested for measurement of enhancer activity at P0, P7, or P28 by amplicon sequencing.
