High-density gene expression profile of 14,151+ unique genes for 6 normal porcine neutrophild and 6 porcine neutrophils samples infected by Salmonella using deep sequencing technology. Peripheral blood samples were collected from 20 healthy crossbreed (Duroc × Landrace × Yorkshire) piglets from a commercial swine farm in Jingzhou, China without sacrificing the animals (ten males and ten females, four weeks old). Prior to blood extraction, the feces were tested negative for Salmonella three times. Neutrophils were isolated through density gradient centrifugation using Ficoll-PaqueTM media (GE Healthcare Life Sciences, Shanghai) as per manufacturer instructions. Luria broth and M9 minimal medium were used to culture Salmonella bacteria. The neutrophils were treated with 106 colony forming units (cfu) of Salmonella enterica serovar Typhimurium (ATCC® 14028™) as previously described [7]. Samples were collected at zero- or eight-hours post infection (hpi). Six samples from each group (zero and eight hpi) were selected for transcriptome analysis.
