This study investigates the epigenetic impact of Nicotinamide N-methyltransferase (NNMT) overexpression on human umbilical vein endothelial cells (HUVECs). HUVECs were transfected with adenovirus vectors, Ad-NC (negative control) or Ad-NNMT (overexpressing NNMT), for 48 hours. Subsequently, protein was collected for chromatin immunoprecipitation analysis using antibodies against H3K4me3 and H3K27me3 via Cut&Tag technology. To further elucidate the effects on DNA methylation, whole-genome methylation sequencing was performed. The resulting data provide insight into NNMT's regulatory role in histone modifications and methylation patterns. Overall design: Human umbilical vein endothelial cells (HUVECs) were cultured and transfected with either Ad-NC or Ad-NNMT for 48 hours. Following transfection, chromatin-associated proteins were subjected to Cut&Tag analysis using H3K4me3 and H3K27me3 antibodies to examine histone modifications. Subsequently, DNA methylation sequencing was performed to investigate the effects of NNMT overexpression on the methylation landscape. Comparative analyses were carried out between Ad-NC and Ad-NNMT transfected cells to understand the role of NNMT in epigenetic regulation.
