G1 Phase Shortening via Chir-124 Treatment in HEK293 Cells

The goal of this experiment was to test the potential of the Chk1 inhibitor Chir-124 to accelerate HEK293 through the G1 phase of their cell cycle. Cells were treated with Chir-124 for 48 hours before being harvested and stained with propidium iodide for determination of percentage of the cell population in each cell cycle phase. Conclusion: Analysis of HEK293 cells following Chir-124 treatment for 48 hours shows substantial loss of cells in the G1 phase, instead showing more even distributions between G1, S, and G2/M phases. Additionally, there is a population of PI stained cells with less fluorescence than the G1 population that may indicate mirconuclei forming with treatment.
