A strictly anaerobic, Gram-staining-positive, spore-forming rod-shaped bacterium, and designated JN-1, was isolated from pit mud. The strain grew at 20-45°C (optimum, 30°C) and pH 5.0-7.0 (optimum pH 6) and in the presence of 0–1.5% (w/v) NaCl (optimum, 0%). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain JN-1 clearly belonged to the genus Clostridium of the family Clostridiaceae. Strain JN-1 shared the highest 16S rRNA gene sequence similarity to Clostridium carboxidivorans DSM15243T (94.21 %), Clostridium scatologenes DSM757T (94.07 %), Clostridium drakei DSM12750T (94 %) and Clostridium acidisoli DSM12555T (94.43 %). The predominant cellular fatty acids of strain JN-1 were C16:0 (47.17%), C14:0 (12.04 %), iso C 15:1 H/13:0 3-OH (10.15 %); The genomic DNA G+C content was 31.17 mol%. Acetate was mainly produced from Glucose, methionine, yeast extract, sorbitol, galactose and serine. Based on the genotypic, chemotaxonomic and phenotypic analyses, strain JN-1 represents a novel species of the genus Clostridium, for which the name Clostridium sp. nov. is proposed. The type strain is JN-1.
