获取胚胎发育期第12,14周的视网膜组织,将组织剪碎成小块,用木瓜蛋白酶和胶原酶IV以及DNase1将其消化成单细胞悬液,终止消化反应后进行流式分选,获取活性较好的视网膜单细胞,利用10x genomics对单细胞悬液进行扩增建库,获取单细胞文库,文库质检合格后送至测序公司使用Illumina平台测序,测序长度为双端150bp。

Retinal tissues at embryonic developmental stages of 12 and 14 weeks were collected. The tissues were minced into small fragments, then digested into a single-cell suspension using papain, collagenase IV and DNase I. After terminating the digestion reaction, flow cytometry sorting was performed to obtain retinal single cells with good viability. The single-cell suspension was amplified and library constructed using 10x Genomics to acquire single-cell libraries. After passing the quality inspection of the libraries, the qualified samples were sent to a sequencing company for sequencing on the Illumina platform, with a read length of paired-end 150 bp.
