Dynamic direct cDNA transcriptome sequencing of equid alphaherpesvirus 1 (EHV-1). Analysis of dynamic transcriptome of equid alphaherpesvirus 1 (EHV-1)

Equine Herpesvirus-1 was propagated on Rabbit Kidney cell line (RK-13/35) using 0.1 MOI (multiplicity of infection). Samples were isolated in different time points as follows: 1h, 2h, 4h, 6h, 8h, 12h, 24h, 48h post infection. Poly(A)-selected RNA was isolated by Macherey Nagel RNA Nucleospin kit and dcDNA library was prepared with the SQK-DCS109 kit. Viral and host cell transcriptome was sequenced on Oxford Nanopore MinION device. The POD5 files were processed into fastq files using the Dorado basecaller. The uploaded gziped files are .fastq files containing viral and host reads. The aim of the study was to detect new viral isoforms during the course of the infection. This dataset provides new insights into the kinetic changes of gene expression of EHV-1 transcriptome as well as the host-virus transcriptomic changes.
