We performed CRISPR/Cas 9 knockout of exon5 and exon7 of the HOIL-1L gene in C2C12 cells and found that impaired myotube maturation occurred. We studied the effect of HOIL-1L knockout on transcriptional changes of C2C12 cells. Â Expression profiling showed that genes involved in muscle contraction and myogenesis were downregulated in HOIL-1L-exon5 and exon7 KO cells compared to controls. Overall design: RNA-sequencing was performed at day 5 of myotube differentiation to assess expression alterations in HOIL-1L-exon5 and exon7 KO C2C12 cells.
