We report that the decreased expression of mitochondrial genes we observe in hepatocyte-specific Nampt knockout mice is normalized in primary hepatocytes, and that hepatocyte isolations causes major changes to the hepatocyte transcriptome for both knockouts and wildtype mice. Overall design: We isolated primary hepatocytes from wildtype and hepatocyte-specific Nampt knockout mice, and sequenced RNA from 1. a liver biopsy following perfusion with saline (L), 2. from washed hepatocytes prior to seeding (CS) and 3. from primary hepatocytes following 24h of culturing (PH) for both genotypes and compared transcriptomes
