High throughput sequencing of a DNA segment of the gene eGFP integrated in the plasmid in Escherichia coli. The gene was mutated by a mutant plasmid (MP) containing a fusion protein of cytidine deaminase and T7 RNA polymerase (T7 RNAP) in E. coli. Compared with controls, the sequencing data presented here demonstrate that MP can be used for in vivo diversification of the target gene in vivo to accomplish directed evolution.
